If you drain a WB combi & then run the hot water will it work properly?

no. water will run but it can't get hot because the boiler won't work while its empty

But, if you turn off the isolation valves on the boiler flow & return before you drain so the boiler is still pressurised then it will work.

I would not do that, you are relying on a 10mm bypass valve inside the boiler, if things went wrong and the boiler left the building via the roof, Who's gonna pay for a new one ? Its not worth the risk,
