Is it possible to use a full bore lever valve to isolate the cylinder coil on the flow side instead of using a motorised valve?

yes it is as long as the flow can ' by pass' through the rads, make sure there is at least one rad without a thermostatic valve or, you will damage the pump because it has no where to push the water,  |
